Thông tin chung: Bệnh lao vẫn là mối đe dọa lớn cho sức khoẻ cộng đồng. Các nghiên cứu cho thấy axit béo chuỗi trung bình (MCFA), như những chất có trong dầu dừa nguyên chất (Virgin Coconut Oil - VCO), có khả năng chống lại nhiều vi sinh vật, kể cả vi khuẩn lao (mycobacterium tuberculosis). Mục đích của nghiên cứu này là xác định độ đáp ứng trong phòng thí nghiệm của vi khuẩn lao với hai nhãn dầu dừa nguyên chất thương mại chứa lượng axit lauric khác nhau và được sản xuất bằng hai quá trình công nghệ khác nhau. Hoạt tính chống lao của hai loại dầu dừa nguyên chất này được thử nghiệm và so với axit lauric nguyên chất cũng như với mẫu đối chứng âm tính. Thí nghiệm được lặp lại hai lần. Mẫu dầu dừa được đã được tiêm vào 0,1 ml vi trùng lao chủng H37RV và ủ ở nhiệt độ 37oC, lắc trong 5 ngày. Sau khi ủ, hỗn hợp được pha loãng thành 4 mẫu, 10 lần một mẫu, đến nồng độ 10-4 và đếm khuẩn lạc (countable colonies) trong môi trường tấm thạch Middlebrook 7H10 và môi trường phân lập vi khuẩn lao khe Lowenstein-Jensen (L-J). Sự phát triển của vi khuẩn lao được kiểm tra hàng tuần trong vòng từ ba đến tám tuần.

Kết quả: Cả hai nhãn hiệu VCO với độ loãng ngày càng tăng từ 1:10 đến 1:10000 cho kết quả đếm khuẩn lạc của vi khuẩn lao giảm từ 46% đến 100% trên thạch Middlebrook 7H10. Kết quả tương tự ghi nhận trên khe L-J với số lượng khuẩn lạc giảm 25% đến 96%. Dung dịch axit lauric nguyên chất ức chế hoàn toàn phát triển của vi khuẩn trong cả tấm thạch và khe L-J.

Kết luận: Các chế phẩm dầu dừa nguyên chất chứa lượng axit lauric khác nhau đã thể hiện mức độ ức chế khác nhau đối với vi trùng lao. Kết quả nghiên cứu cho thấy tiềm năng của dầu dừa nguyên chất như tác nhân chống lại phát triển của vi trùng lao. Cần nghiên cứu tác động tương tự đối với các sinh vật có hai khác. Cần nhiều nghiên cứu sâu hơn, sử dụng dầu dừa nguyên chất với các nồng độ khác nhau cũng như tác dụng của với các thành phần khác nhau như axit lauric và các axit béo khác trong dầu dừa như axit myristic, axit caproic hay axit caprylic.

ABSTRACT:

Background: Tuberculosis remains a major public health threat. Studies have shown that medium chain fatty acids (MCFA), such as, those found in Virgin Coconut Oil (VCO), possess activity against a wide range of microorganisms including Mycobacterium tuberculosis. The goal of the study was to determine the in-vitro susceptibility of M. tuberculosis (TB) isolates to two commercial brands of VCOs each containing different amounts of lauric acid and produced by two different extraction processes. Two brands of VCO were tested for their anti-TB actions compared to pure lauric acid and a negative control. Two replicate runs were done. VCO samples were inoculated with 0.1 mL of the H37RV strain of M. tuberculosis and incubated at 370C with shaking for 5 days. After incubation, the mixtures were diluted 10-fold up to 10-4 concentration to obtain countable colonies using Middlebrook 7H10 agar plates and LowensteinJensen (L-J) slants. Colony growths of M. tuberculosis were examined in each dilution every week for three to eight weeks.

Results: Both brands of VCO at increasing dilution from 1:10 to 1:10000 lowered the colony counts of M. tuberculosis by 46% to 100 % on the Middlebrook 7H10 agar. Similar results were noted on L-J slants with colony counts decreasing by 25% to 96%. Lauric acid solution showed complete inhibition of bacterial growth in both the agar plates and LJ slants.

Conclusion: Different VCO preparations containing different amounts of lauric acid exhibited different degree of inhibition against M. tuberculosis. The result of the study suggests the

But more studies are needed to replicate the studies using different concentrations of VCOs, lauric acid and other fatty acids in VCO such as myristic acid, caproic or caprylic acids.
